Study Identifies 3 Candidate Biomarkers Associated With T-Cell Infiltration in Psoriasis
Three genes associated with immune cell infiltration may represent promising diagnostic biomarkers and future therapeutic targets in psoriasis, according to a study examining the molecular mechanisms underlying T cell–driven inflammation.
Psoriasis is a chronic inflammatory skin disease characterized by dysregulated immune responses, with T cells playing a central role in disease pathogenesis. To better understand the molecular basis of immune infiltration, investigators performed a comprehensive analysis to identify biomarkers associated with T cell activity in psoriasis.
The analysis identified 3 candidate biomarkers—AKR1B10, C10orf99, and CKS2—that demonstrated strong diagnostic performance. According to the investigators, all 3 biomarkers exhibited “high sensitivity and specificity” in receiver operating characteristic analyses, and the diagnostic nomogram developed using these markers demonstrated reliability.
Gene set enrichment analysis further suggested that the identified biomarkers participate in immune-related signaling pathways. The 3 genes showed significant enrichment within the NOD-like receptor signaling pathway and focal adhesion, both of which have been implicated in inflammatory responses.
Immune infiltration analysis revealed increased levels of activated B cells and CD8 T cells in psoriasis tissue compared with control samples. The 3 biomarkers also demonstrated meaningful correlations with multiple immune cell populations, suggesting that they may reflect broader immune activity within psoriatic lesions.
To support the bioinformatic findings, investigators performed preliminary reverse transcription quantitative polymerase chain reaction validation. This analysis confirmed increased expression of AKR1B10, C10orf99, and CKS2 in psoriasis tissue samples, consistent with the computational results.
According to the authors, “AKR1B10, C10orf99, and CKS2 may serve as candidate molecules for future mechanistic studies” and may provide “potential diagnostic biomarkers for further investigation of psoriasis-related immune regulation.”
