KRAScendo-1: Divarasib Significantly Improves Survival Over Sotorasib or Adagrasib in KRAS G12C-Mutated Non-Small Cell Lung Cancer

Clinical Summary:

  • Ferdinandos Skoulidis, MD, PhD, discusses results from the randomized phase 3 KRAScendo-1 trial comparing divarasib with sotorasib or adagrasib in patients with previously treated locally advanced or metastatic KRAS G12C-mutated non-small cell lung cancer (NSCLC).
  • Divarasib significantly improved progression-free survival, overall survival, and objective response rates compared with sotorasib or adagrasib, with fewer high-grade treatment-related adverse events and treatment discontinuations.
  • These findings support divarasib as a potential new treatment standard for previously treated KRAS G12C-mutated NSCLC, demonstrating superior efficacy with manageable toxicity compared with first-generation KRAS G12C inhibitors.

Ferdinandos Skoulidis, MD, PhD, MD Anderson Cancer Center, Houston, Texas, discusses results from the randomized phase 3 KRAScendo-1 trial comparing the next-generation KRAS G12C inhibitor divarasib with sotorasib or adagrasib in patients with previously treated locally advanced or metastatic KRAS G12C-mutated non-small cell lung cancer (NSCLC).

Dr Skoulidis reviews the significant progression-free and overall survival benefits observed with divarasib, including improved response rates and durable antitumor activity across clinical and molecular subgroups. He also discusses the manageable safety profile, reduced rates of severe treatment-related adverse events, and the potential for divarasib to establish a new standard of care in previously treated KRAS G12C-mutated NSCLC.
